FhuA also serves as transporter of the antibiotic albomycin, which is structurally
related to ferrichroe (figure 1). FhuA also actively transports the structurally unrelated rifamycin CGP 4832, thereby rendering E. coli 200-fold more sensitive to CGP 4832 than to the unmodified rifamycin (rifampicin). The crystal structures of FhuA with albomycin (figure 2) and with CGP 4832 have been determined (unpublished data) [11]. Both antibiotics bind mostly to the same amino acid residues that bind ferrichrome. Unexpectedly, albomycin assumes two rather different conformations, an extended and a bent form, both of which are tolerated by the surface cavity of FhuA [11]. All four sites in which CGP 4832 differs from rifamycin (figure 1) serve to bind CGP 4832 to FhuA. This may explain why FhuA serves as an active transporter for CGP 4832 but not for rifamycin.
