Rabies virus is a single-strand, negative-sense RNA virusof the family Rhabdoviridae. Its nonsegmented genomicRNA acts as a template for both transcription and replication.During the transcription process, a positive-strandleader RNA and five monocistronic mRNAs are synthesized,whereas during the RNA replication process, a full length,positive-strand RNA (replicative intermediate) is synthesized,which serves as the template for the synthesis ofprogeny negative-strand genomic RNA (Wunner, 1991). Theprogeny genomic RNA is encapsidated with nucleoprotein(N) and this RNA–N complex, together with the nominalviral phosphoprotein (P, also designated NS or M1) andRNA polymerase (L), forms the active cytoplasmic ribonucleoprotein(RNP) complex in virus-infected cells and theRNP core in virus particles (Sokol et al., 1971). The RNPcomplex alone is capable of initiating viral RNA transcriptionand replication in vivo and in vitro. Recent studies ofrabies viral protein functions using reverse genetics demonstratedthat N, P, and L in the RNP complex are allrequired for rabies virus RNA transcription and replication(Conzelmann and Schnell 1994; Schnell et al., 1994). However,the sequence of events, specificity, and mode(s) ofinteraction between each of these proteins and betweenthese proteins and the genomic RNA which results in viralRNA transcription and replication has not been defined
